"We will not under any circumstances have a second referendum", Downing Street said.
Johnson said "Britain stands on the brink of the greatest crisis since the Second World War".
"But I have every hope that the Prime Minister would listen to Parliament, because the alternative in those circumstances would be nearly certainly that the Government would fall". The "democratic thing to do is to give the public the final say", he argued.
The timing of the decision will come as a blow to Mrs May, who is priming her Cabinet to sign up to proposals which will allow her to secure a deal on the withdrawal agreement with Brussels.
"Britain stands on the brink of the greatest crisis since the Second World War", said Johnson, a former Financial Times journalist who voted to stay in the European Union in the 2016 referendum.
The deal being negotiated with Europe by the government - dubbed the "Chequers Deal" - would leave Britain economically weakened with no say in European Union rules it would have to adhere to for years, while a "no deal" scenario would inflict damage on the country.
The only alternative on the table, he said, is a no-deal Brexit that would "inflict untold damage" on Britain.
He acknowledged that the Brexit negotiations "have at least united us in fraternal dismay".
He told a tech industry meeting in London on Wednesday: "I don't think it is a question so much of the risk of major shortages, but I think probably the average consumer might not be aware of the full extent to which the choice of goods that we have in the stores are dependent on one or two very specific trade routes".
Lib Dem leader Sir Vince Cable, whose party supports calls for a "People's Vote" on the final deal, said: "We warmly welcome Jo Johnson's support of the campaign to give the people the final say on the deal and a chance to exit from Brexit".
"In fact so great is the gulf now between what was promised in the referendum campaign and what is now on offer in the Prime Minister's proposed deal that I have had no choice but to submit my resignation to the Prime Minister this afternoon".
Labour Shadow Brexit Minister Jenny Chapman said that Theresa May had "lost all authority and is incapable of negotiating a Brexit deal within her own party, let alone with the EU".
